Oxford City Council suspended The Imperial nightclub's licence following a hearing last month.
A deadline passed at midnight on Tuesday for the club's managers to appeal.
The club, in Park End Street, has not appealed and must now close until Wednesday, May 21.
The council's licensing committee made the suspension order after police compiled a dossier of trouble at the club including drugs and drink-fuelled violence, including toilets testing positive for drug and a mass brawl.
They blamed a series of disturbances on cheap drinks promotions.
Tony Cope, Thames Valley Police licensing officer, said officers will speak to the licensee when the suspension period is over to discuss ways to improve the club's record.
The Imperial nightclub stayed open for three weeks during which time it could appeal against the suspension.
It can apply to host events during the six week licence suspension, which will be reviewed individually by the council and police.
